Zhang F, Feridooni T, Hotchkiss A, Pasumarthi KB. Divergent cell cycle kinetics of midgestation ventricular cells entail a higher engraftment efficiency after cell transplantation. Am J Physiol Cell Physiol 308: C220–C228, 2015. First published November 19, 2014; doi:10.1152/ajpcell.00319.2014.—Cardiac progenitor cells (CPCs) in the primary and secondary heart fields contribute to the formation of all major cell types in the mammalian heart. While some CPCs remain undifferentiated in midgestation and postnatal hearts, very little is known about their proliferation and differentiation potential. In this study, using an Nkx2.5 cell lineage-restricted reporter mouse model, we provide evidence that Nkx2.5 CPCs and cardiomyocytes can be readily distinguished from nonmyocyte population using a combination of Nkx2.5 and sarcomeric myosin staining of dispersed ventricular cell preparations. Assessment of cell number and G1/S transit rates during ventricular development indicates that the proliferative capacity of Nkx2.5 cell lineage gradually decreases despite a progressive increase in Nkx2.5 cell number. Notably, midgestation ventricles (E11.5) contain a larger number of CPCs ( 2-fold) compared with E14.5 ventricles, and the embryonic CPCs retain cardiomyogenic differentiation potential. The proliferation rates are consistently higher in embryonic CPCs compared with myocyte population in both E11.5 and E14.5 ventricles. Results from two independent cell transplantation models revealed that E11.5 ventricular cells with a higher percentage of proliferating CPCs can form larger grafts compared with E14.5 ventricular cells. Furthermore, transplantation of embryonic ventricular cells did not cause any undesirable side effects such as arrhythmias. These data underscore the benefits of donor cell developmental staging in myocardial repair.

Relation between Age, Gender, Number of Blood Products Received and Diagnosis with Engraftment and Length of Hospital Stay in Patients Undergoing Autologous Stem Cell Transplantation

Background and purpose: Autologous hematopoietic stem cell transplantation may be one of the best treatment options for some patients with hematological malignancies. The process of stem cell homing in the bone marrow and the production and proliferation of denovo cell populations is called engraftment and could be considered as one of the key factors in the success of stem cell transplantation...

Association between CRP Level on Admission Day and Length of Hospitalization and Engraftment in Patients Undergoing Autologous Hematopoietic Stem Cell Transplantation

Background and purpose: Several studies investigated the role of C-reactive protein (CRP) during transplantation and inflammation, and clinical outcomes after stem cell transplantation. In this study, we examined the relationship between the earliest CRP level on admission day, and the first outcome after transplantation, myeloid and platelet engraftment, and the length of hospitalization in p...

Bone marrow stromal cells and their application in neural injuries

Background: This article reviews experimental and clinical studies in which neural injuries repaired with bone marrow stromal cells. History: Bone marrow contains two kinds of stem cells: hematopoietic and nonhematopoietic (stromal) stem cell. In vitro studies indicate that bone marrow stromal cells have the capacity of differentiation into other cells (such as neural cell) under treatment wit...

Co-Transplantation of VEGF-Expressing Human Embryonic Stem Cell Derived Mesenchymal Stem Cells to Enhance Islet Revascularization in Diabetic Nude Mice

Background: Pancreatic islet transplantation has emerged as a promising treatment for type I diabetes. However, its efficacy is severely hampered due to poor islet engraftment and revascularization, which have been resulted to partially loss of transplanted islets. It has been shown that local delivery of vascular endothelial growth factor (VEGF) could accelerate transplanted islet revasculari...
